Nearly 30 years have passed since Koopa was defeated, and everything seemed to be in peace. Luigi Mario runs the plumbing business alone ever since his brother, Mario was taken from him. But after a mysterious message from the fungus, Luigi is led to believe his brother is still alive and is in the fungus. So Luigi and some other people return to the other dimension to survive through the fungus war and search for Mario.
